Abstract:
      Immunoaffinity column(IAC) for T-2 toxin was prepared with anti-T-2 toxin monoclonal antibody 2G7 and CNBr activated Sepharose 4B.The experimental conditions for T-2 immunoaffinity column were optimized and the columns were applied in cereals and feed pretreatment.An IAC/LC-MS/MS method was established for the determination of T-2 toxin in cereals and feed.The calibration curve for T-2 toxin standard solution was linear in the range of 0.5-500.0 ng/g with correlation coefficient of 0.999 7.The limit of detection(LOD) and limit of quantitation(LOQ) was 0.05 ng/g and 0.17 ng/g,respectively.The average recoveries of T-2 toxin in rice,maize and feed samples at three spiked levels of 10,50,100 ng/g ranged from 92.9% to 109.7%,with relative standard deviations(RSDs) of 2.1%-8.3%.The method was applied in the detection of T-2 toxin in 36 real samples with RSDs less than 10.0%.
